The production of CAR-expressing T cells (CAR-T cells) requires a workflow incorporating cell culture, cell sorting, genetic engineering, and gene transfer techniques. Briefly, blood is extracted from a donor, from which T cells are extracted using cell processors or cell separators. The T cells are then placed into cell culture and activated using antigen presenting cells (APCs) or antibodies. At this point, the genetic material encoding the CAR is introduced to the activated T cells, and T cells expressing the CAR are selected, expanded, and then delivered to a recipient or placed into storage.1
