Beckman Coulter's Particle Characterization (PCC) portfolio contains several products specialized in counting and analyzing different types of cells and particles. An overview of the PCC products is presented highlighting the correlation between the particle size and quality. Furthermore, the Coulter principle was explained underlining the cell Volume as a more sensitive parameter of cell size change which cannot be detected by imaging or laser diffraction methods.

Flow cytometry is widely used to analyze extracellular vesicles (EVs), including investigation of their potential as biomarkers of disease. Unfortunately, conventional flow cytometry has many limitations, primarily because of the wide range of EV sizes, and the inability of most flow cytometers to detect particles < 0.5 μm. In this 40-minute video, Dr. Radu explains how new generation flow cytometry technology—such as the CytoFLEX platform—can count, analyze and characterize EVs of widely variable sizes. In addition, CytoFLEX cytometers can reduce inter-laboratory variability in the detection of EV size.

Analysing and using tissue with haematological malignancies are pivotal for understanding disease prognosis, progression, mechanisms, and its potential treatment. However, variability among patients and samples hinder research outcome times. In order to speed up research discoveries, the parallel use of analyser systems (Cytoflex LX) and sorters (Cytoflex SRT) for numerous samples will help standardise disease progression while allowing to perform experiments at multiple timepoints. The new MRC National Mouse Genetics Network in the UK will allow to have multiple disease models where mutations could be timed and will allow to monitor disease progression. An ex-ante and ex-post-facto situation of several samples will allow to discern the mechanisms in health and malignancy. The presentation “Hematopoietic stem cell panel transfer from analyser to CytoFLEX SRT cell sorter" introduces the field of haematology regarding hematopoietic stem cells how its output might be affected in malignancy. The presentation concludes with showing how to use the analyser/sorter in haematology and why it is important for this field.

The CytoFLEX LX cytometer has been pivotal to research by African and UK partners working in Sudan, Ethiopia, Kenya and Uganda to better understand the pathogenesis of leishmaniasis and the underlying immune mechanisms. A consortium is working in parallel to clinical trials of a candidate vaccine to prevent chronic dermal leishmaniasis in patients treated for visceral leishmaniasis. The promising vaccine has been developed by Professor Paul Kaye and his team at the University of York, United Kingdom. This 39-minute video provides details of the team’s fascinating clinical and research journey, including some of the real-world challenges they encountered.

Flow cytometry is a quick way to evaluate the number of metabolically active cells—and to distinguish between viable and dead microorganisms—which is extremely important for the probiotic and food industry. Regardless, many companies in this industry still use labor- and time-consuming quality control analytical techniques based on plating samples onto media and counting the resulting colonies after incubation. The results of cytometric enumerations of bacteria and yeasts based on different staining protocols, in comparison to classical microbiology methods, are presented in this 42-minute video.

One crucial cellular subset in the tumor microenvironment is represented by myeloid-derived suppressor cells (MDSC), which have marked immunosuppressive and pro-tumorigenic capabilities. MDSC are composed of immature and mature cells capable of inhibiting both innate and adaptive immune responses. Previous studies have shown that human MDSC could be detected in different tumors, and are capable of interfering with T- and NK-cell proliferation and/or function. The presence of MDSC at the tumor site—or in the peripheral blood of tumor patients—has been associated with a poor prognosis. In this 33-minute video, Dr. Tumino explains how MDSC could represent a promising new target to enhance clinical outcomes for patients.
